Öğe Effects of subclinical pregnancy toxemia and L-carnitine supplementation on angiogenesis, energy metabolism, and fetal development in ewes(Elsevier B.V., 2026) Turgut, Ali Osman; Bilen, Ebru Karakaya; Koca, Davut; İşbilir, Fatma; Özöner, Özgür; Ünver, Ali; Şirin, Muhammed HasanSubclinical pregnancy toxemia (SPT) is a common metabolic disorder affecting small ruminants during late gestation under conditions of negative energy balance. This study investigated the effects of experimentally induced SPT on angiogenesis-related gene expression in uterine and placental tissues, hepatic energy metabolism-related gene expression, fetal and placental development, and selected serum biochemical parameters in ewes. The potential protective effects of L-carnitine supplementation were also evaluated. Nineteen crossbred Hamdani ewes were assigned to three groups: SPT (G1, n = 6), SPT supplemented with L-carnitine (G2, n = 7), and control (G3, n = 6). SPT was induced by dietary restriction from gestational day 100. Blood samples were collected on days 100, 110, 120, 130, and 138, and tissues were collected at day 138 for morphometric, histopathological, and molecular analyses. Maternal body weight, uterine weight, fetal weight, and placentome dimensions were significantly lower in G1 than in controls (p < 0.05). Histopathology revealed significant hepatic lipid accumulation in G1. Serum β-hydroxybutyrate, triglyceride, and cholesterol concentrations increased, whereas glucose concentrations decreased in G1 during late gestation (p < 0.05). L-carnitine supplementation improved these metabolic alterations in G2. Placental expression of VEGF, VEGFR1, and VEGFR2 was significantly downregulated in G1 compared with G2 and G3 (p < 0.05). Hepatic DGAT1 expression increased significantly in G1 (p < 0.05), whereas GPD1 and GK expression remained unchanged. These findings indicate that SPT impairs fetal and placental development, disrupts angiogenic signaling, and alters hepatic lipid metabolism. Dietary L-carnitine supplementation may compensate these adverse effects and improve maternal metabolic status and placental function in ewes. © 2026 Elsevier B.V.Öğe Immunohistochemical demonstration of Toll Like Receptors - 2, 4 and 6 in mycoplasmal pneumonia(Ankara Üniversitesi, 2026) Yücel, Gözde; Tunç, Arda Selin; Özöner, Özgür; Ulusoy, Yavuz; Kılınç, Bahadır; Vural, SevilMycoplasmal pneumonia is a highly contagious disease that causes serious economic losses in sheep, goats, and cattle worldwide. The aim of this study was to investigate the pathomorphological findings and the immunohistochemical distribution of Toll-Like Receptors (TLRs) involved in the pathogenesis of mycoplasmal pneumonia (n=40). Histopathological examination revealed, purulent bronchopneumonia (n=13), fibrinous bronchopneumonia (n=14), interstitial pneumonia (n=6), granulomatous pneumonia (n=1), and non-specific lesions (n=6). The same tissue sections were immunohistochemically stained with antibodies against Mycoplasma pneumoniae, TLR-2, TLR-4, and TLR-6. Immunopositivity for TLRs was observed in bronchial, bronchiolar, alveolar epithelial cells and inflammatory cells. Although no statistically significant difference was found between the groups, TLR-2-positive cell counts were numerically higher in purulent inflammation, whereas TLR-4-positive cell counts were numerically higher in fibrinous inflammation. The results indicate that TLR-2, TLR-4, and TLR-6 may contribute to the inflammatory response in mycoplasmal pneumonia.Öğe Spontan Gelişen Köpek Pyometralarında Aquaporinlerin Rolü ve Ovaryum Steroidleri, Antioksidan Enzim Aktiviteleri ile İlişkisinin Ortaya Konulması(2023) Yazlık, Murat Onur; Vural, Mehmet Rıfat; Vural, Sevil; Tunç, Arda Selin; Müştak, İnci Başak; Kaya, Ufuk; Özöner, ÖzgürPyometra, köpeklerde diöstrus döneminde gözlenen genellikle ölümle seyreden yangısal bir hastalıktır. On yaşından daha genç köpeklerin ortalama %19?unda gözlenebilmektedir. Genlerin amplifikasyonunda SYBR Green I boya içeren kit kullanılmıştır. Virülens genlerin belirlenmesi amacıyla konvansiyonel PCR yöntemi kullanıldı. Serum progesteron, LPS, SOD, GPx ve NO düzeyleri antikoagulansız tüplere alınan kan örneklerinin santrifüjü sonrası elde edilen serumdan ölçülmüştür. Doku ve serumdaki antioksidan aktivite, toksin düzeyleri ve serum progesteron değerleri ticari ELİSA kitleri kullanılarak belirlenmiştir. Pyometra olgularında ovaryum, uterus ve serviks dokusunda AQP ekspresyon seviyelerinin değişim gösterdiği; pyometraların açık veya kapalı serviks tipinde olmasına göre uterus dokusunda AQP ekspresyonlarının farklılaştığı; izole edilen E.coli?lerde virülens genlerin bulunması durumunda AQP?lerin uterus dokusundaki ekspresyonlarında farklılıklar gözlenebileceği; pyometralı köpeklerde kanda artan LPS düzeyinin uterus AQP ekspresyonlarıyla ilişkili olduğu; köpeklerde pyometra olgularında serviksin açılma mekanizmasında AQP?lerin rol alabileceği; SOD, GPx ve NO düzeylerinin uterus ve serviks dokusundaki bazı AQP ekspresyonlarından ile ilişkili olduğu belirlenmiştir.
