Öğe Efficacy of Punica granatum L. and Matricaria chamomilla L. herbal extracts on wound healing(Univ Zulia, Facultad Ciencias Veterinarias, 2026) Polat, Eren; Ates, Asiye Ozlem; Ozkan, Ahmet Selim; Kaya, Emre; Ozoner, Ozgur; Pekince-Ozoner, Merve; Serin, HakanDermal injuries, which account for a significant portion of healthcare expenditures globally, occur in various forms due to the influence of various etiological factors, primarily trauma. For all these reasons, wound healing is among the most researched topics worldwide. This study aimed to investigate the effects of ointments obtained from Punica granatum L. (pomegranate) and Matricaria chamomilla L. (chamomile) extracts on secondary wound healing. 42 Sprague Dawley rats were used in the study. In the study, a total of six experimental groups were formed, each consisting of 7 rats: control group, healthy control group, Punica granatum treatment group, Matricaria chamomilla treatment group, Punica granatum and Matricaria chamomilla combination group (Punica granatumtreatment + Matricaria chamomilla treatment), and the group receiving an application containing olive oil, beeswax, and natural vitamin E. An excisional wound model was created using a biopsy punch on the back of the subjects in all groups except the healthy control group. While ointments prepared from herbal extracts were applied to the rats in the Punica granatum treatment, Punica granatum treatment + Matricaria chamomilla treatment, and Matricaria chamomilla treatment groups, rats in the control group received 0.9 % NaCl, and rats in the olive oil, beeswax, and natural vitamin E group were treated with a mixture of olive oil, beeswax, and vitamin E without any herbal extracts. At the end of the study, it was determined that the Matricaria chamomilla treatment group had the highest mean in terms of wound closure percentage and wound perimeter change percentage, while the Punica granatum treatment group had the lowest mean (P < 0.05). Although the best groups in terms of histopathological scoring were the Punica granatum treatment and Matricaria chamomilla treatment groups, no statistical difference was observed between the groups (P > 0.05). In terms of oxidative stress parameters (except for superoxide dismutase), the treatment groups were statistically significantly closer to the healthy control group than to the control group. Considering all evaluation criteria, it was concluded that Matricaria chamomilla plant was more effective on wound healing than Punica granatum plant and was a more suitable option for clinical use.Öğe Efficacy of water-based propolis in preventing intra-abdominal adhesions in an experimental rat model(Univ Zulia, Facultad Ciencias Veterinarias, 2026) Polat, Eren; Sakin, Dilan Selin; Cimen, Irmak; Kaya, Emre; Can, Batuhan; Ozoner, Ozgur; Pekince-Ozoner, MerveIntra-abdominal adhesions are one of the most common postoperative complications following surgical procedures. Globally, it is among the leading causes of hospital readmissions, chronic pain, and serious conditions such as infertility in both human and veterinary medicine. The risk of development is particularly high after pelvic and abdominal surgeries. Despite this common condition, there is still no effective and reliable treatment method to prevent adhesions. This study aims to evaluate the potential effect of propolis (both oral and intra-abdominal) with its anti-inflammatory and antioxidant properties in the prevention of intra-abdominal adhesions. The rats used in the study were divided into five: Healthy Control Group , Control Group , Sham Group , Oral Propolis Group , and Intra-abdominal Propolis Group . After the intra-abdominal adhesion model was established in rats, excluding the Healthy Control Group and Sham Group, rats in the Oral Propolis Group were administered 75 mgkg-1 of propolis orally once daily for 20 days, while rats in the Intra-abdominal Propolis Group received a single dose of 75 mgkg-1 of propolis intra-abdominally. Results showed that the Oral Propolis Group and Intra-abdominal Propolis Group were statistically significantly lower than the Control Group in terms of macroscopic adhesion scores. Although there was no statistically significant difference when the Oral Propolis Group and Intra-abdominal Propolis Group were compared with the Control Group in terms of fibrosis, it was observed that the Intra-abdominal Propolis Group was statistically significantly lower than the Control Group in terms of inflammation severity. Biochemical analyses, it was revealed that the oxidative stress parameters of rats in both oral and intra-abdominal propolis application were statistically different from the Control Group and that these two were close to the Healthy Control Group in terms of oxidative stress. As a result, it was concluded that both intra-abdominal and oral propolis applications prevent the formation of intra-abdominal adhesions and can be used for prophylactic purposes after abdominal and pelvic operations.Öğe Expression patterns and distribution of aquaporin water channels in cervix as a possible mechanism for cervical patency in bitches affected by pyometra(Elsevier Science Inc, 2024) Yazlik, Murat Onur; Ozkan, Hueseyin; Vural, Sevil Atalay; Kaya, Ufuk; Ozoner, Ozgur; Mutluer, Ipek; Altinbas, Yunus FurkanPyometra is a life-threatening disease, the severity of which depends on cervical patency status. This study investigated cervical inflammation status as well as the expression patterns and localization of aquaporin (AQP1, AQP2, AQP3, AQP5, and AQP9), and hormone receptors in cervical tissue that influences canine pyometra. Of the 36 animals enrolled in the study, 24 were diagnosed with pyometra and separated into two groups: open cervix pyometra and close cervix pyometra, while 12 healthy animals presented for elective ovariohysterectomies were allocated into the control group. Surgical treatment was performed for treatment of pyometra. After each operation, cervix samples were collected and analyzed for AQP and hormone receptor expression patterns determined by qPCR and protein expression by means of immunohistochemistry. Blood samples were also collected to determine serum progesterone concentrations. AQP9 expression was downregulated approximately 3-fold while and PGR expression was downregulated more than 2 fold in both pyometra groups compared to the control group. AQP3 and AQP5 gene expression levels were upregulated more than 3 fold in the open-cervix pyometra group than the closed-cervix pyometra group (P < 0.05). This is the first study to describe the expression patterns and immunolocalization of AQPs in canine cervical tissue based on pyometra patency status and to report AQP3 and AQP5 expression in cervical tissue linked to cervical patency.Öğe Morphometric, macroscopic and microscopic investigation of glandulae uropygiale in turkeys (Meleagris gallopavo)(Wiley, 2024) Isbilir, Fatma; Ozguden Akkoc, Cansel Guzin; Isbilir, Ihsan; Ozoner, Ozgur; Guzel, Baris Can; Arican, Ilker; Atalgin, Sukru HakanThis study was carried out to investigate the macroanatomical, morphometric, histological, and electron microscopic characteristics of the uropygial gland in adult male and female turkeys (Meleagris gallopavo). The glandula uropygialis of 13 adult turkeys (6 males and 7 females) were used for this study. It was determined that this glandular structure located dorsal to the last caudal vertebrae was heart-shaped and consisted of two lobes and a papilla system. As a result of statistical evaluation, significant differences were found between the sexes in the parameters of lobe length, papilla width and papilla height (p < .05). Also, papilla uropygialis index (PUI) value had a statistically significant difference (p < .05). Tissue samples taken to determine the histological structure of the gland were stained with Hematoxylin & Eosin, Crossman's triple, periodic acid Schiff (PAS), diastase, and orcein. Histological examination revealed that the gland had a two-lobed structure surrounded by a capsule composed of connective tissue. It was determined that the gland had a tubular structure and the epithelial layer consisted of cellular layers as a germinative layer, intermediate layer, secretory layer, and degenerative layer from the periphery to center. In the scanning electron microscope examination, the lobe structure of the gland consisted of different shaped follicles protruding from the surface. It was observed that these glandular follicle structures were gathered together in different ways or mostly disorganized. As a result of the study, it was determined that the morphological and histological structure of the uropygial gland in turkeys was similar to that of other bird species, but showed some species-specific and habitat-dependent differences in general. Ar & imath;can Research Highlights The uropygial gland consisted of a system of two lobes and one papilla in each of male and female turkeys. The glandular lobes consisted of follicles surrounded by connective tissue, while the follicles were connected to each other by interfollicular septae. The results obtained in morphometric measurements revealed statistical differences between male and female birds. Histological examination showed periodic acid Schiff (PAS) (+) reaction in the basal membranes of tubule epithelial cells and secretion. Lymphoid cell communities were found in both connective tissue and intertubular regions. The fact that the reaction was not glycogen-induced was determined by the observation of PAS positivity as a result of staining with the PAS-diastase technique.Öğe Mycobacterial infection in a Nile crocodile (Crocodylus niloticus) from Türkiye(Ankara Univ, 2024) Ahlat, Ozan; Ozoner, Ozgur; Filikci, Kursat; Vural, Sevil AtalayMycobacterial infection in Nile crocodile tissues sent from a private zoo was characterized pathomorphologically and immunohistochemically in this case. Macroscopically, multifocal, greyish -white areas ranging in size from 1 mm to 5 mm were seen in the lung, liver, and spleen. Histologically, a large number of well -demarcated necrotic areas were seen. These areas included nuclei debris locally. Inflammatory cells along with a couple of multinucleated giant cells surrounded the necrotic cores. Numerous acid -fast bacilli were detected by Ziehl-Neelsen staining method. Immunolabelling for both Mycobacterium bovis and anti-BCG antibodies was positive in each tissue.Öğe Protective effect of dietary L-carnitine supplementation on follicular development in the ovary of ewes with subclinical pregnancy toxemia(Copernicus Gesellschaft Mbh, 2026) Turgut, Ali Osman; Pekince Ozoner, Merve; Koca, Davut; Karakaya Bilen, Ebru; Isbilir, Fatma; Ozoner, Ozgur; Isbilir, IhsanThis study examined the effects of subclinical pregnancy toxemia (SPT) on follicular development and evaluated the protective potential of dietary L-carnitine supplementation in ewes. Eighteen crossbred Hamdani ewes underwent estrous synchronization, natural mating, and pregnancy confirmation via ultrasonography on day 45 post mating. The ewes were fed according to the National Research Council (NRC) (2007) guidelines until day 100 of gestation, after which they were assigned to one of three groups: SPT (G1, n=6), SPT+L-carnitine (G2, n=6), and control (G3, n=6). Blood beta-hydroxybutyrate (beta HBA) levels were measured on days 100 and 138. Following slaughter, ovarian tissues were collected, processed, stained with Harris hematoxylin and eosin, and imaged for histometric analysis. Follicle types, oocyte diameters, and zona pellucida (ZP) thickness were recorded. Primordial follicle numbers did not differ significantly among groups (p>0.05). Primary follicles tended to be higher in G1 compared to G2 and G3, but this was not statistically significant (p<0.05). Secondary follicles differed significantly, with G2 exhibiting higher counts than G1 and G3 (p<0.05), indicating enhanced secondary follicle development with L-carnitine supplementation. Antral follicles were low across all groups, with no significant differences (p>0.05), consistent with limited preovulatory follicle formation during pregnancy. Atretic follicles were more numerous in G1 than in G2 and G3 (p<0.05), suggesting increased follicular regression in ewes with subclinical pregnancy toxemia (SPT). Secondary follicle oocyte diameters were larger in G2 and G3 than in G1 (p<0.05), while antral follicle oocyte diameters showed no significant differences (p>0.05). ZP thickness tended to be higher in G3 for both secondary and antral follicles, although differences were not significant between groups (p>0.05). Pairwise effect sizes (Cohen's d) also indicated a large effect of L-carnitine on secondary follicle and antral follicle development, and oocyte diameter; and a negative effect of subclinical pregnancy toxemia on follicle development. These results highlight the importance of adequate nutrition and metabolic support during late gestation and suggest that L-carnitine supplementation may enhance follicular growth and reproductive performance in ewes with SPT. Further studies are needed to clarify the mechanisms by which L-carnitine protects ovarian functions and follicular development.Öğe Protective effects of BAIBA and thymoquinone in type 1 diabetic nephropathy: modulation of Irisin, NF-?B, and Caspase-3 expression(Springer, 2026) Pekince Ozoner, Merve; Gur, Fatih Mehmet; Aktas, Ibrahim; Ozoner, Ozgur; Timurkaan, SemaDiabetes is closely related to increased production of reactive oxygen species, which leads to oxidative stress, chronic inflammation, and increased apoptosis, especially in kidney tissues. Irisin is a recently discovered myokine with the potential to offer hope for the treatment of many metabolic diseases, while BAIBA is also a newly identified endogenous protective myokine. In this study, the effects of thymoquinone (TIM), known for its antioxidant activity, as well as the possible agonistic interaction between irisin and BAIBA on cellular stress and apoptosis occurring in diabetic kidneys were investigated using immunohistochemical methods. In this study, 35 Sprague Dawley rats were equally separated into five groups: Control, STZ, TIM, BAIBA and STZ + TIM + BAIBA. Type 1 diabetes was induced by a single intraperitoneal injection of streptozotocin (STZ, 50 mg/kg). The same protocol was applied to induce diabetes in the TIM and BAIBA groups. Following induction, TIM (20 mg/kg) and BAIBA (100 mg/kg) were administered daily via gavage for five weeks. In the STZ + TIM + BAIBA group, diabetes was induced similarly, followed by daily oral administration of a combination of TIM and BAIBA at the same doses for five weeks. At the conclusion of the study, kidney samples were obtained and analysed using both histochemical and immunohistochemical methods. Results demonstrated that TIM significantly reduced intersitial fibrosis by 55% in the kidneys. It is revealed that both TIM and BAIBA reduced NF-kappa B immunointensity by 63% and when used simultaneously by %48. Caspase3 immunointensity was reduced by 38%, 46% and 26% following TIM, BAIBA and TIM + BAIBA administration respectively. Also both TIM and BAIBA was observed to cause positive up-regulation on irisin expression. The findings of this study demonstrated that TIM and BAIBA effectively prevented renal fibrosis and apoptosis in STZ-induced diabetic rats, particularly through the downregulation of NF-kappa B.Öğe Regulatory effects of PNX-14 on BPAinduced abnormalities in puberty, steroidogenesis, and reproductive histopathology in male and female rats(Wiley, 2026) Akipek, Mehmet Emre; Ercan, Eda Coban; Akipek, Rumeysa Esra Kom; Atmaca, Ahmet Tuluhan; Ozoner, Merve Pekince; Ozoner, Ozgur; Canpolat, Sinan[Abstract Not Available]Öğe Seasonal investigation of the macroscopic and microscopic structure of the sinus interdigitalis in Hamdani crossbred sheep (Ovis aries)(Wiley, 2024) Isbilir, Fatma; Ozguden Akkoc, Cansel Guzin; Kirman, Gunsel; Ozoner, Ozgur; Ozoner, Merve Pekince; Isbilir, Ihsan; Gulendag, ErmanBackground: The odours of different body parts and structures, such as external genitalia, female urine, ventral tail radix, infraorbital sinus and interdigital sinus, have a strong attractiveness for rams. The sinus interdigitalis is considered an important trail gland for sexual behaviour, as its odour is included in the list of strongly attractive structures for male animals during the breeding season, being important in pheromone production. Objectives: In the present study, the effects of sex and breeding season on the morphological and histological structure of the interdigital gland were investigated in Hamdani crossbred sheep (Ovis aries). Methods: Sinus interdigitalis glands located on the forelimbs and hind limbs were collected from 10 male and 10 female sheep during both breeding and non-breeding seasons. The gland's position in the forelimbs and hind limbs was determined radiographically. The glands were examined macroscopically, histologically and immunohistochemically in both seasons. Results: In all animals, the topographic location of the gland between the digits was determined to be distal to the phalanx proximalis and at the level of the phalanx media. The mean weight of the gland in male and female animals was 1.07 +/- 0.03 and 1.4 +/- 0.11 g, respectively. In general, the results of morphometric measurements were higher during the breeding season, and the increase was more pronounced in female animals. In histological examinations, it was determined that both neck and body parts of the tissues consisted of a structure consisting of a connective tissue capsule at the outermost, dermis, epidermis and lumen underneath. Immunohistochemically staining revealed that oestrogen receptors showed positive immunoreactions only in the intracytoplasmic localization of apocrine sweat glands. In terms of morphological, histological and oestrogen receptors, it was thought that the gland may play a role in sexual communication.Öğe The presence of virulence factor genes downregulates uterine AQP3 and alters glutathione peroxidase activity and uterine histopathology in canine pyometra(Wiley, 2024) Yazlik, Murat Onur; Mustak, Inci Basak; Ozkan, Hueseyin; Vural, Sevil Atalay; Kaya, Ufuk; Ozoner, Ozgur; Mutluer, IpekPresent study was designed to evaluate the role of virulence factor genes (papG, cnf1 and hylA) in the pathogenesis of canine pyometra. Antimicrobial susceptibility test and detection of virulence genes were performed Escherichia coli (E. coli) detected in uterine swab samples. Animals were divided into two groups based on the presence (VF+, n:14) or absence (VF-, n:7) of the virulence factor genes papG, cnf1 and hylA. Blood and tissue glutathione peroxidase activity, uterine histopathologic analysis and AQP3, ESR1, PGR, OXTR gene expressions were determined in both groups. Statistical analyses were performed using Stata version 15.1. All E. coli isolates were susceptible to amikacin, whereas resistant to ampicillin, amoxicillin/clavulanic acid and lincomycin. None of the isolates were susceptible to cefotaxime. E. coli isolates had at least one virulence gene. The most prevalent gene was fimH (100%), followed by fyuA (95.8%), usp (83.3%), sfa (75%), cnf1 and hlyA (70.8%) genes. Blood GPx activity was greater in VF+ animals. On the other hand, uterine tissue GPx activity was lower in VF+ group compared to the control group. Expression levels of AQP3 were upregulated more than fivefold in VF-dogs compared to the control group. In addition, AQP3 expression levels were found approximately threefold higher in VF (-) than VF (+) group (p < .05). Varying degree of inflammation noted for all animals with pyometra, but the presence of bacteria noted only in VF+ animals. In conclusion, the presence of virulence factor genes does not play a role in the histopathological degree of inflammation, the presence of bacteria was found to vary. Serum GPx activity increased in VF+ animals. While the hormone receptor expressions were similar, AQP expression was upregulated in the absence of virulence factor genes.
