In this study, the presence of Bt8, Bt9, Bt10, and Bt11 genes associated with resistance to common bunt disease (Tilletia spp.) was investigated using molecular markers in a total of 70 bread wheat landraces collected from different agro-ecological regions. PCR-based molecular analyses were performed to determine the resistance genes, and the obtained results were evaluated by gel electrophoresis. The marker Xgwm114 was used for the detection of Bt8, Bt10, and Bt11 genes, Xgpw7433 for the detection of the Bt9 gene, and FDA/RSA was used to confirm the presence of the Bt10 gene. The results revealed that the majority of the examined genotypes did not possess markers associated with the resistance genes evaluated in this study, indicating that a significant proportion of the landraces may be potentially susceptible to common bunt disease. Among the investigated genes, Bt10 was the most frequently detected, whereas Bt9 was identified at the lowest frequency. The Bt8 gene was detected in a limited number of genotypes, while the Bt11 gene was found at a very low frequency. Furthermore, no genotype carrying all four resistance genes (Bt8, Bt9, Bt10, and Bt11) simultaneously was identified. Genotype-based evaluations revealed that the Bt8 gene was mainly concentrated in Erzurum "Kırik" and Konya "Kamçı" genotypes, whereas the Bt10 gene was more common in genotypes originating from Gümüşhane, Hakkâri, and Konya. The Bt11 gene was detected only in genotypes from Konya and Hakkâri, while the Bt9 gene, has not been detected in any genotype. The presence of multiple resistance genes in Hakkâri genotypes suggests that these materials may possess a higher and more durable resistance potential against common bunt disease. As a result, landraces carrying resistance genes may serve as important genetic resources for breeding programs aimed at developing common bunt-resistant wheat cultivars.
